Yes, it would work.
No, it would not work as well as epoxy.
Polyester resin works fine for all-glass layups, but it doesn't have the adhesive properties that epoxies have. Polyester resin is also somewhat permeable, so water can eventually--not soon, but eventually--seep in behind the glass and ruin your glass-wood bond and the boat could fail. This may take years to happen, but you won't get an heirloom kayak that you could build with epoxies.
If you're looking at polyester resins to save costs, look at Raka epoxies. Their resin systems are nearly as inexpensive as polyesters, but much, much stronger and better for wood boat construction.
If you're looking at building with polyester just for the learning experience of building with polyester, then by all means, build with poly, and let us know how it holds up in a few years. I've heard of polyester/glass wood boats that have lasted 15-20 years. I've also heard of glass that you could peel off by hand after as little of two seasons' use.
