What you are describing is essentially the same as a bung or spigot, the design of which has long been used in sealing containers of liquid beverages.

You could take the simple route of buying collapsible 2 to 5 gallon water containers and inflating them.

Several table wines are available in boxes, the liners of which are easily recycled into use as flotation. I suggest you keep several handy at the launching of your new craft. Should you have any need of assistance in emptying such containers, send me an e-mail. I'm always happy to help kayak builders. :)

Oh, chemical supply, and hospital supply firms have lots of designs for pinchcocks and hose clamps in metal and plastic. Certainly you can find some that will close your tubing easily and cheaply.
