With due respect to Bill, no the beam does not have to be straight.
You can adjust the forms on the beam to get the forms straight, if you have enough clearance between the form and the beam to make adjustment.
However, I had a problem with a 2x4 plywood beam in that when supported at 2 points it bent enough that I could not get the forms adjusted enough.
I eventually added a 3rd center support, then angled braces closer to the end of the beam before I got enough of the sag out. The hull was on top to start, so when the hull was stripped (with staples and glued) the entire thing was stiff enough to hold the shape when I turned it over in a couple of cradles.
That boat was done with string alignment and I had serious issues with getting the frames properly position.
Next boat I borrowed a laser level and got it done very quickly and accurately. My wife bought me one for Christmas and it is well worth the expense (I am sure some will think me anal). But you have to have a vertical centerline and and the same horizontal waterline on all forms (they would normally be there, but the boat I built was designed for a ladder and only had the waterline below the keel - off the forms)

Every tiny bit of height over 2x4 beam will reduce your issues with the beam sagging. Additionally making the upper and lower elements of the "2x4" beam of solid wood instead of plywood will help also. The stiffer the wood the better. This is not the place for cedar.
